  8.2.1
Sample Collection & Handling
1. Collect upper respiratory swab sample per applicable clinical guidelines. Nasopharyngeal and oropharyngeal swabs may be tested individually or combined in the same transport medium tube. The following transport media are compatible with the test:

􏰀 3ml Copan UTM + Copan Flocked Swab (Copan, Cat. No. 305C)
􏰀 3ml Puritan UniTranz + Flocked Swab PurFlock Ultra (Puritan Medical

􏰀 2ml Virocult + mini foam swab (Medical Wire, MW950SENT)
3ml transport medium volumes are recommended where possible. Small swabs suitable for nasopharyngeal sampling should be used.
Swab samples in transport media can be stored at 2-8oC for up to 48 hours prior to testing.
2. Close the cap of the vial containing the transport medium and swab and vortex for 10 secs. It is recommended to centrifuge briefly to ensure all liquid is at the bottom of the tube.
Instructions for Use
8.2.2 Extraction
Extract 140μl of the sample per the “Purification of Viral RNA (Spin Protocol)” instructions (including all recommended spin steps) in the RNA Mini Kit IFU, with the following critical exception: at the final step, elute nucleic acid from the column by adding 140μl of COVID dilution buffer (COVID-DB) without enhancer instead of the 60 μl Buffer AVE supplied with the QIAamp kit. After extraction proceed to assay set up per section 8.4.
