Applications of spectrofluorimetry:
I- Spectrofluorimetric determination of Quinine sulfate
The techniques of spectrofluorimetry and phosphorimetry measure the intensity of
the light emitted from a system that has absorbed radiant energy.
Preparation of stock solution:
Prepare a 100.0 µg/mL quinine sulfate stock solution by quantitatively transferring
exactly 10 mg quinine sulfate to 100.0 mL volumetric flask, adding 5 mL 1 M
sulfuric acid, and diluting to volume with deionized water.
This stock solution was further diluted with 0.05 M sulfuric acid as appropriate to
obtain the working concentration range.
Procedures:
1) Transfer aliquots of the working standard solution into a series of 10 mL
volumetric flasks.
2) Complete to the volume with 0.05 M sulfuric acid.
3) Measure the fluorescence intensity at 455 nm after excitation at 350 nm, using
0.05 M H 2SO 4 as a blank.
