B) Streaking on agar slant or agar plate:
By means of a sterile loop, take an inoculum from the culture tube.
Streak on an agar slant beginning from the bottom of the tube & moving upwards, from side to
side, without scratching the agar surface.
Incubate at 37oC for 24 hrs.
Examine visually for microbial growth.
C) Stabbing in agar deep:
Using a straight loop “stab”, transfer a small volume of the liquid culture into the agar deep using
the stabbing technique demonstrated.
Examine visually for microbial growth “turbid lines”.
