III: SUBCULTURING OF BACTERIA & FUNGI
Aim: To carryout sub-culturing of bacteria and fungus
Methods of cultivation of microorganisms:
1 cell cultivation 1 colony
A) Transfer into broth = subculture or inoculation
Culture in a liquid medium is known as inoculation. It is used as refreshment of the organism, by
growing it in a fresh medium.
Using the aseptic technique, inoculate a loopful of the organism into the clear sterile nutrient
broth tube.
Incubate at 37oC for 24 hrs.
Examine visually for microbial growth “turbidity”.
