Factors affecting Drug retention
• Lipid structure:
– Lipid (S) chain length: increasing the chain length
increases the transition temperature of the system so?
– Lipid unsaturation reduces the transition temperature of
the system so?
• Cholesterol content:
– Cholesterol reduces the bilayer permeability of the
liposomes as I reduces he mobility of the phospholipids
within the bilayer.
– Improves the in vivo stabiliy of liposomes by reducing the
loss of phospholipids to lipoproteins
– N.B. cholesterol content shouldn’t exceed 50 mol%, why?
