Steps:

1- Preparation of different dilutions of the test disinfectant and the phenol in the

five empty sterile test tubes for the test disinfectant prepare (1/1000,1/1100,1/ 1200

and 1/ 1400) and for the phenol prepare (1/90, 1/100, 1/105, 1/110, 1 115).

2- Preparation of the reaction mixtures to the prepared dilutions transfer 0.2 ml of

the microorganism at zero time to all prepared dilutions at 30 seconds interval, then

make the reaction mixtures stand for 2.5, 5, 7.5 and 10 minutes.

3- Making subcultures from the reaction mixtures

Divide the nutrient broth tubes into four sets (four time sets 2.5, 5, 7.5, and 10

minutes sets, each set composed of five n. broth tubes), then take one loopful from

each reaction mixture and inoculate it to the corresponding nutrient broth tube in

each time set at 30 seconds interval.

3- Incubate the nutrient broth tubes at 48-72 hours at 37 ̊C.
