Synergism Examples
1) Sequential inhibition of successive steps in metabolism
• e.g. sulphonamide- trimethoprim
• Treatment and prevention of chronic urinary tract infection, typhoid fever
and shigellosis.
2) Prevention of emergence of resistant populations
e.g. combination therapy for TB.
3) Inhibition of inactivating enzymes
e.g. Aminopenicillin/ -lactamase inhibitor combinations
Ampicillin-sulbactam, Amoxicillin-clavulanate
Extended-spectrum Penicillin/ -Lactamase inhibitor Combinations
Piperacillin-tazobactam, Ticarcillin-clavulanate.
• These compounds act as suicide inactivators of the bacterial beta- lactamases
enzymes
