6 Flame the neck of the test tube.

7 Insert the loop into the culture broth and
withdraw.
Note: You don’t need to be able to see any
globs of bacteria on the loop! There are
plenty there by simply touching the surface
once.

8 -Flame neck of the test tube and replace the lid/cotton wool plug on the test tube

using the little finger.
-Place test tube on bench.
10 Partially lift the lid of the Petri dish
containing the solid medium.

11 Hold the charged loop parallel with the

surface of the agar; smear the inoculum
backwards and forwards across a small area
of the medium to create the first streaks
