EFFICACY
COMPARISON OF CYTOTOXIC EFFECTS ON RABBIT
CORNEAL ENDOTHELIUM BETWEEN PRESERVATIVE-FREE
AND PRESERVATIVE-CONTAINING DORZOLAMIDE/ TIMOLOL

ABSTRACT
PURPOSE: To evaluate and compare the toxic effects of eyedrops containing a fixed combination of
2.0% dorzolamide and 0.5% maleate timolol with or without preservatives on rabbit corneal endo-
thelium.
METHODS: In vivo study on 11 rabbits comparing Dorzolamide/Timolol eyedrops with preservative
or without preservative. The left eye was treated with BAK DTFC while the right eye was treated with
preservative free DTFC: eyedrops were injected into the anterior chamber. Corneal endothelium toxic
damages are evaluated after 24 hours.
STUDY CRITERIA:
X Clinical exam: corneal haze and conjunctival and limbal vascular injection were
graded from 0 (none) to 4 (severe).
X TUNEL: detection of apoptotic cells (cell death).
RESULTS:
X Slit lamp photograph shows 24 hours after injection of BAK DTFC and PF DTFC :
X A: BAK DTFC. The eye show severe corneal haze (2.8± 0.6) and conjunctival vascular
injection (2.4± 0.8)
X B: PF DTFC. The eye show minimal corneal haze (0.4± 0.5) and conjunctival vascular
injection (0.6±0.5)
