DAY 2-4
The DNA Typing kit is a flexible kit, with several stopping points for teachers who have short class
periods or slow gel electrophoresis apparatuses. The following instructions will assist you in
determining how to organize this kit for your class.
MELTING AND CASTING GELS
May be performed in the early part of the class period; for long classes, you may begin to run the gels.
Otherwise, store the gels in a ziploc bag overnight in the refrigerator.
RUNNING THE GEL
Add the BlueVis running buffer to the electrophoresis chamber when you plan to start running the gel
do not allow the gel to set overnight in the buffer. For classes with slower electrophoresis boxes (where
the gel will take longer than the class period to run), optional InstaStain sheets have been included to
resolve the DNA so that the gel may be stored overnight.
ANALYZING THE GEL IN REAL-TIME
While the gel is running, and for several hours after it has run, the DNA bands may be viewed with the
BlueVis dye present. BlueVis dye will fade with time, and cannot be stored overnight.
ANALYZING THE GEL WITH INSTASTAIN
If the gel must be stored overnight, or you desire to resolve the bands more for photodocumentation
purposes, you may restain the gel with the included InstaStain sheets following the procedure below:
1. Place the gel in a shallow plastic container filled with distilled water.
2. Float the InstaStain sheet, blue side down, on the water.
3. Allow the gel to stain and destain overnight.
