                                    QUALITY CONTROL OF ROSEBENGAL ANTIGEN AND

                                             VALIDATION OF ROSEBENGAL TEST

                           Economic  impact  of  brucellosis  is  extensive  and  ranked  among  the  most
                     important zoonosis globally due to posing great damage in livestock productivity and

                     human health. From diagnostic point of view, rose bengal test (RBT) is most common

                     and effective screening test with low cost and high sensitivity.
                           Although  ‘Biocombinate’  STATE  OWNED  company  of  Mongolia  have  been

                     producing  rose  bengal  antigen  (RBA)  since  1978  for  the  diagnosis  of  livestock
                     brucellosis, evaluation and validation of RBT utilising local reference sera and RBA

                     produced  in  many  countries  still  have  not  been  done.  Thus,  we  have  aimed  to
                     conduct  comparative  evaluation  of  diagnostic  capacities  of  RBA  produced  at

                     Biocombinate to RBA produced at ID Vet of France using local reference sera. Prior

                     to this, we have examined cultivation and dissimilation of B. abortus 99 strain used in
                     the production of RBA in 9 different selective and basal culture medium such as meat

                     peptone  agar,  meat  peptone  broth,  Trypcase  soya  agar,  Tryptone  soya  agar,

                     Brucella  agar,  Blood  agar,  Agar  with  Thionine,  Streptomycin  and  Penicillin,
                     respectively.  Biochemical  characteristics  and  motility  was  identified  in  liquid  and

                     semi-liquid  medium  and  haemolytic  feature  was  checked  by  selective  “Вlood  agar
                     base” medium.

                           General  characteristics  of  the  B.  abortus  99  strain  are  confirmed  to  be
                     conserved  which  suggested  in  strain  passport  of  Biocombinate  SOSSE  and World

                     Organisation  for  Animal  Health  (OIE)  listed  requirements.  Furthermore,  we  have

                     examined the B. abortus 99 strain by Bruce-Ladder polymerase chain reaction (PCR)
                     which  was  recommended  by  World  Animal  Health  Organization  (OIE)  and  the  B.

                     abortus 99  strain  was  confirmed to  be  not underwent for  any  genetic mutations  or
                     deletions. Along with this, quality control tests were inspected according to Veterinary

                     Pharmacopeia-2009 on final product of RBA and quality passed RBA was utilized for
                     our further examination.
