Evolution of HPLC

❑ In traditional column technology, particle size of the
   stationary phase was fairly large (>100 µm).

❑ Looking for higher resolution, smaller particle size
   stationary phase packing material was developed (<10
   µm).

❑ Fine column packing (3-50 μm) required high pressure
    pumping systems capable of pushing the mobile
    phase through the column at pressures up to 6000 psi
    to achieve appropriate flow rates.
