Equipments:
1. Two flasks, one containing 100 ml of 5% phenol (Standard Disinfectant) and the
other one containing 100 ml of 2% test disinfectant.
2. Flask containing sterile distilled water.
3. Flask containing inoculated yeast suspension (2ml of 24h broth culture + 48ml
of 3% yeast suspension). Yeast is used as a source of organic matter.
4. 4 tubes (sterile & empty).
5. 4 n. broth tubes (containing 10 ml).
6. 10 ml pipette.
7. Loop.
Steps:
1- Preparation of four different dilutions of both the phenol & the test
disinfectant in the four empty sterile test tubes Ex: for phenol:
Take 10ml from phenol flask to first empty test tube transfer 2.5ml & reject 7.5
ml. so after this step the first empty test tube will contain 2.5ml of 5%phenol and
the phenol flask will contain 90 ml of 5% phenol.
Then, transfer 10ml sterile distilled water to the flask of phenol so its
concentration will be 4.5%.
