Microbial Enumeration Test
Microbial enumeration test is a quantitative test which
determines the Total Aerobic Microbial Count (TAMC) and
Total Yeast and Mold Count (TYMC) present in the test product.
• Total Aerobic Microbial Count (TAMC): Number of
colony forming units (cfu) found using Tryptic Soy Agar (TSA).
• Total Combined Yeasts and Molds Count (TYMC): Number of cfu found
using Sabouraud- Dextrose Agar (SDA).
• The total viable aerobic count is the sum of the bacterial count and the fungal
count
Microbial Growth Agar Media
Soybean-Casein Digest Agar- for TAMC
Sabouraud’s Dextrose Agar - for TYMC
• The test product is dissolved or diluted (1:10 dilution) in Phosphate Buffer
Solution (pH 7.2) or Soybean–Casein Digest Broth.
• A surface-active agent (such as polysorbate 80 may be added to assist the
suspension of poorly wettable substances.
Techniques
1-Plate Count methods
• Pour-Plate Method
• Surface-Spread Method
2- Membrane Filtration method: Uses filtration apparatus
• Plate Count Methods— Perform plate count methods at least in duplicate for each
medium and use the mean count of the result.
