Pour Plate Method
You are provided with a pharmaceutical product, determine the Total Aerobic
Microbial Count (TAMC) and the Total Combined Yeasts and Molds Count
(TYMC).
Equipment
1. Test product
2. Two Test tubes of diluent
3. Flak of Soybean-Casein Digest Agar
4. Flask of Sabouraud-Dextrose Agar
5. Two 1ml pipettes
6. 4 petri dishes
Method
1. Using a glass marker, mark diluent test tubes as 1: 10, 1: 100 dilutions
2. Using the sterile pipette, transfer 1 ml of test product to the first test tube
containing 9 ml diluent (1:10), mix thoroughly.
3. Using the same pipette, transfer 1ml from the first dilution to the second
diluent tube (1:100).
4. Mark the plates 1: 10, 1:100
5. Transfer 1ml of each dilution to the center of the corresponding petri dish.
6. Add 15 to 20 ml of melted media that maintained at not more than 45 ̊C to
petri dishes.
(soybean–casein digest agar to two petri dishes and Sabouraud-dextrose
agar to the other petri dishes).
7. Mix gently by rotating the plate clockwise and anticlockwise, back and
forward.
