Identification of MC4R gene makers in local goats with

Abstract. The melanocortin 4 receptor (MC4R) gene is a gene that affects sympathetic nerve
activity, adrenal and thyroid function, and as a medium for leptin to work in regulating energy
balance and homeostasis. This study aims to identify the diversity of the MC4R gene in Bligon
goats. This research was conducted in the Gama Ngundi Lestari livestock group using 10 pre-
weaning bligon goats with single and twin birth types. The research material used is recording
data (birth weight and weaning weight data). This research has two stages. The first is blood
sampling and data analysis. Two DNA polymorphism of the MC4R gene were identified in the
exon region (g.998A / G and g.1079C / T). The analysis was carried out by calculating genetic
diversity using the Hardy-Weinberg formula. The present study of MC4R gene in Bligon goat
detected non-synonymous polymorphism in location g.998A/G. Which was changed amino acid
methionine (Met) to isoleucine (Ile) and the SNP at g.1079C/T was a synonymous
polymorphism. Restriction enzyme KpnI and BsrI may be used for genotyping Bligon goat
using PCR-RFLP method in the future research.
