Let’s use the Michaelis-Menten equation to figure out Km and Vmax!

v = Vmax [S]0                                             Let’s do it!
     [S]0 + Km

                              1
If we take the reciprocal v , we get...

1 = [S]0 + Km = [S]0 + Km
v Vmax [S]0 Vmax [S]0 Vmax [S]0

                  = 1 + Km × 1
                    Vmax Vmax [S]0

In other words, we can draw the graph of the straight line y = ax + b, where y : 1
                                                                                             v

y x: 1
                                                                                    [S]0

                         Slope a         a : Km
                                            Vmax

                                         b: 1
                                            Vmax

                         x

 A linear function? I That’s right! The graph
learned about those of a linear function is
in middle school. No just a straight line, so
                            this makes our job much
  problem! (I think...)
                                   easier because...

                                     No matter how far
                                     we extend the line,
                                       our formula will

                                           always work!
