Procedure

1. Dilute 100 μl of sugar solution with 10 ml distilled water

2. Add 20 μl sample + 2.0 ml reagent,

20 μl standard + 2.0 ml reagent,

2 ml reagent (blank)

3. Mix and incubate 10 minutes at 37 C or 30 minutes at room temp.; the
color is stable 30 minutes at room temp.

4. Read the absorbance of the sample (Ab sample) and standard (Ab
standard) against reagent blank at wavelength λmax 505 nm (490-550
nm).

Calculations

• Standard concentration = 100 mg%

• Dilution Factor = 100
