Procedure

1- Apply 0.1ml extract onto a silica gel plate, along side a reference

sample of khellin.

2- Develop the plate with EtOAc (Ethyl acetate).

3- Locate the khellin band using UV at 366 nm.

4- Scrape khellin band using thin spatula, quantitively transfer to a

small funnel.

5- Elute khellin by 4ml boiling water, add 5ml 2N H 2SO 4 to the cooled

khellin solution and complete the volume to 10ml with water, leave

for 5 min.

6- Measure A at 420 nm using blank.

Notes

- H2SO4 is added to increase the intensity of the yellow color

to be measured colorimetry.
