ABSTRACT
Cryopreservationtechniqueisveryesentialinthevariousfieldssuchasfodandmedicalenginering. Especialy,inthecaseofplant,preservationwithlivingisdesiredunderlowtemperature.However,mostof plantmaysufersomedamageduetocoldstresunderlowtemperature.Theauthorshavecontinuedto investigatethedamagedecreaseofplantunderlowtemperaれlre.Consequently,wefoundthatthemicro electriccurentloadcausesthedamagedecreaseefectonplanttisueandleaf.However,becausethereisno establishedestimationfortransientleafdamageatpresent,weightchangeratioandmicroscopicobservation betwenbeforeandaftertheexposuretolowtemper.qtureweretemporaryusedinourpreviousresearch.Then, normalized diference vegetation index (hereafter,NDVI) taking use of spectrum information in photosynthesiswasapliedinaditiontotheabovemeasures.However,NDVIwasalwaysnotasatisfactory transientmeasure.However,企omdepinspectionofthesespectrumresults,wenoticedthattherearethe characteristicchangesinwavelengthfrom500nmto630nmwiththeprogresofcoling.Onthebasisofthis spectrumcharacteristicchange,thisresearchproposedthenewmeasure(index)toestimatetransientleaf damagewithhigheracuracyanddiscusedtheposibilityofnewindex.Tounderstandleafdamage,chemical substancesrelatedtophotosynthesis;chlorophylaandscarotene,wasmeasuredunderlowtemperatureby HPLC.Consequently,itwasfoundthatnewindexwasmoresuitablethanNDVIforestimationofleafdamage.

Cryopreservationtechniqueisveryesentialinthevariousfieldssuchasfodandmedicalenginering. However,astypicalbiomaterialcontainsalotofwater,waterspontaneouslychangestoiceoutsideorinside ofcelthroughsupercoledstatebelow企ezingpointwiththeprogresofcoling.Itisgeneralysaidthat intracelularfrezingmaycausecel'sfataldeath.Especialy,mostofplantsaredesiredtobepreservedwith living.Therefore,cryopreservationtechniqueforplantissaidtobeverydificult.Thereweremanypaperson plantdamageundercoldstres.Alabove,Yoshida'sgrouphasactivelyperformedtheinvestigationonlow temperatureaclimationofplant[1].However,thereisfewreportsfromtheviewpointsofunderstandingof intracelularfrezingbehavioursandthedamagedecreaseofplantthatweareinterestedin. Undertheabovebackground,wediscuseddamagedecreaseefectduetomicroelectriccurentloadtofind theposibilityofnewcryopreservation.Todoit,firstweconstructedthecryo-microscopicsystemequiped withahighspedcamerainaditionordinaryCCDcameraandacoledCCDcamera.Next,usingthis
