Plastic-contaminated cooking oil increased AST and ALT levels and damaged the Liver of Rats
(Rattus norvegicus)

ABSTRACT
Snacks such as fried food in public ever reported contaminated plastic due to mixture. Plastic contamination will
be toxic and free radicals that damage digestive organs like the liver. The injury of the liver gives a sign through
microstructure description and AST-ALT level. The research purpose was to determine the effect of mixture
cooking oil and plastic in Rats (Rattus norvegicus) through liver histopathology and AST-ALT level. As many as
twenty Rats, male, 150-250 grams, 8-12 weeks divided four groups consist of a negative control group, and
treatment groups 0.5 mL/day, 1.0 mL/day, 1.5 mL/day for seven days respectively. The mixing was 0.5 kg plastic
PP (Poly Propylene) blended hot frying oil 1L administrated orally via gastric lavage. The euthanization of rats was
conducted on the 8 day via blood exsanguination in anaesthetized rats. Then liver was collected in Formalin

10%. The serum's AST-ALT level was measured, then continued statistic analysis using One Way ANOVA and Post
Hoc Tukey Test 95%. The results showed that plastic and cooking oil mixture increased significantly (p<0.05) for
AST and ALT blood levels. The mixture also impacted the elevation of liver injury related to elevation mixture
volume. The histopathology liver portraited several pathology descriptions, such as inflammation and necrosis in
the tissue. The severity of liver injury escalated based on the elevation of mixture volume. The conclusion is that
the plastic-contaminated cooking oil induces liver tissue damage and elevation of AST-ALT blood levels.
