Blood agar

• Preparation: melt nutrient agar to 100 C, cool to 55 C,

Add 5-10 % sterile blood of sheep or ox, mix well,

poured into plate or slopes and allowed to cool and

solidify.

• Color: red opaque color

• Uses: - As Enriched media

• It differentiates organism according to the

action on blood.

Note: Blood agar is enriched &differential media

Chocolate agar Heated blood agar

• Preparation: it is prepared as blood agar then raise the

temperature to 100 c for 2 mins then pour into plate.

• Color: it is brown chocolate color.

• Uses: for cultivation of haemophilus group of organisms

Lofflers serum

• Composition: ox or horse serum in glucose broth and sterilized in hot air oven at 75

C for 3 hrs on 2 successive days

• Color: opaque white media

• Used for cultivation of Corynbacterium diphteria.
