Steps to prepare stable pH gradient
1. A suitable ampholyte solution is incorporated
into the polyacrylamide gel.
2. On applying an electric field, the ampholytes
separate into zones of defined pH. If the
ampholyte system is well designed, a stable and
smooth pH gradient is established in the gel.
3. The gel with the established pH gradient is
ready now for protein analysis.
