Mobile Phase Selection

           Guidelines for Setting Mobile Phase Conditions

❑ If the sample is cationic i.e. basic:

                Reversed phase mode is still an option…. But

❑ Mobile Phase Strength has to be adjusted by changing Eluent

Composition to be either:

➢ Basic buffer solution / acetonitrile.  What is the
➢ Basic buffer solution / methanol.      problem??

❑ This method is certainly effective with regard to separation,
   but it creates another problem and is not often used.

Increase retention strength by making the eluent alkaline
