HPLC Column

❑ Typical HPLC columns are 10, 15 and 25 cm in length & are

filled with extremely small particles (3, 5 or 10 µm).

Column Types

✓ Narrow bore column (15cm x 2.1mm) <50 µg of sample.
✓ Analytical column (25cm x 4.6mm) <100 µg of sample.
✓ Semi-preparative column (25cm x 10mm) <100 mg.
✓ Preparative column (25cm x 21.2mm) <500 mg.
✓ Guard columns.
✓ Pre-columns.

Decreasing the column internal diameter 158
reduces the sample load, produces less
waste, and increases the back pressure;

thus requires higher pump pressure.
