HPLC Column
Column Oven Analysis is often performed
at 40 C to 60 C.
❑ It is very important for column temperature control.
❑ if the column temperature increases by heating:
✓ The load pressure decreases due to a decrease in the
viscosity of the eluent.
✓ The retention time becomes shorter.
✓ Resolution improves by hastening diffusion of solutes.
Heating Mechanisms
❑ Air circulation heating type is commonly used. 161
❑ Block heating type: Aluminum block heater.
❑ Insulated column jacket type: Water bath.
