Aims of Derivatization
1Increasing volatility (i.e. sugars)by masking polar OH, NH,COOH & SH
groups

2decreasing volatility to allow analysis of very low molecular weight
compounds
3 Increasing thermal stability

4Increasing detectability, (i.e. steroids/ cholesterol )by introduction of
ECD detectable groups, such as halogenated acyl groups

5 facilitate the chromatographic separation

6 Reduction of adsorption of polar samples on the active surfaces of the
column walls and the solid support, hence decrease tailing.
