Penciclovir

Prodrug famciclovir

• The same mechanism as acyclovir (bind to DNA polymerase)

Uses: Penciclovir: has little oral activity,
used topically for cold sores
Famciclovir: orally active for the treatment of
o genital herpes

o shingles.

Penciclovir

Prodrug Valganciclovir

• similar structure to acyclovir

• phosphorylated in infected cells by
✓ viral thymidine kinase……(HSV, HZV)
✓ CMV kinase……..(CMV)

Uses:
• Treatment of cytomegalovirus (CMV) infections in
immunocompromised patients

Because CMV does not produce thymidine kinase
it is not normally susceptible to acyclovir and
