Preparation method of primary cell culture

To grow subculture, the monolayer culture was selected, its nutrient medium was

removed in sterile condition, and cells were washed with phosphate buffer solution 1

or 2 times.

Then cells were peeled off by using 1:9 mixed solutions of 0.02% versin and 0.25%
trypsin, suspended in media with 2% serum and then the suspension was

centrifuged at 1000 rpm for 10 minutes.

Supernatant was removed, the cells were diluted in nutrient medium, and live cell

counts were determined. Then the material was poured into culture flask, incubated

at 37 С for 2 to 3 days, and then the cells were grown as monolayer culture.
