2. Determination
(Bromothymol blue)

SCOPE
To estimate the Urease activity for Soya bean meal by
Bromothymol blue indicator method.

REAGENTS
Preparation of Bromothymol blue solution

F5gm Urea in 100 ml double distilled water
(solution 1) Picture - 1 Picture - 2

F0.05gm Bromothymol blue in 10 ml of ethanol or
propane-2-ol (Solution 2)

FCombine1& 2 Solution. Add 1 ml 0.1N Sulphuric
acid. This solution must be freshly prepared.

PROCEDURE
Weigh1 g of fine ground sample of Soya in a test tube.
Add 10 ml of Bromothymol solution. Shake well set
aside for 10 minutes and observe superannuated Picture - 3 Picture - 4
solution. ( Picture - 1)
