5. Determination of Plant Protein

Carbohydrates from plants contain starch and
cellulose. When it reacts with iodine and chlor-zinc FA purple brown color indicates the presence of
iodine solution, the starchy tissue releases a blue color plant fiber, whereas yellow indicates animal fiber
and the plant fiber or cellulose develops a purple- (protein) using a microscopic examination.

REAGENTS
FIodine solution: 0.5 g I and 1.5 g KI dissolved in 25
ml distilled water.
FChlor-zinc-iodine solution: Dissolve 100 g ZnCl
2
in 60 ml water, and then add 20 g KI and 0.5 g I.
PROCEDURE I

FMix 1-2 g test sample with 100 ml boiling water or
boil the mixture for 2-3 min and filtrate the solution.

FPlace a few ml of the cooled mixture on a spot plate
or test tube and add 5-6 drops of iodine solution. If
starch is present, the mixture turns blue.
