COX-2 inhibition 410

• inhibition of COX-2 that produces the desirable effects of
NSAIDs.

• The relatively selective COX-2 inhibiting ( meloxicam ) was the
first step towards developing a true cox-2 selective inhibitor.

• COX-2 selective inhibitor ,Coxibs, the newest class of
NSAIDs, can be considered as true COX-2 selective
inhibitors, and Include celecoxib, rofecoxib,valdecoxib.

Acetaminophen does also work mainly
by blocking COX-2, but unlike the newly
developed COX-2 inhibitors it has

weaker peripheral inhibitory activity
Experts aren't sure exactly how acetaminophen works, but

suspect it blocks a specific type of cyclo-oxygenase (COX)
enzyme, located mainly in the brain.

FIRST GENERATION NSAIDS:

Salicylates

¨ The most known and widely used of these is
"Aspirin“
e.g.: Aspirin®, Jusprin®, Rivo® and Aggrex®.

¨ Other salicylates include Mg salicylate, Na salicylate
and Choline salicylate.
