Efficiency of Separation

To achieve successful chromatographic analysis, we
    should get :

    sharp separated symmetrical chromatographic peaks
    within a reasonable run time

                                    ↑ rate of band separation
    ↓ rate of band broadening

      Band Broadening & Column Efficiency

    • Column efficiency refers to the amount of band broadening
that occurs when a compound passes through the column

    • ↓ Band broadening → ↑ Column efficiency
    • This means that band broadening must be limited
    • During elution, each solute undergoes thousands of transfers

       between stationary and mobile phase
    • Movement of solute down the column can occur only while
    • the molecule is in the mobile phase

     • It is beneficial to measure the efficiency of the column 16
