Comparative study of Coxiella burnetii founded in two provinces in Java, Indonesia

ABSTRACT
Coxiella burnetii is an obligate intracellular Gram negative bacteria known as causative bacteria of Q fever, a
zoonotic disease. Q fever cases in Indonesia were mainly reported to be found in ruminants in the range of 2006
– 2019 with a coverage area of Jakarta, West Java, Yogyakarta, Bali, and North Sumatra. In this study we compared
different result from previous study in West and East Java. Phylogeny analysis was performed to analyse genetic
relationship between C. burnetii DNA that found in imported cow from West Java and local goat from East Java. C.
burnetii nucleotide sequences of com1 gene from imported cow of West Java and goat of East Java are identical.
Meanwhile, using 16S rRNA gene, only one out of 4 sequence from East Java goat that identic with West Java cow
sequence, even they shared in the same branches. C. burnetii mostly found in heart of imported cow from West
Java, while in local goat of East Java, this bacteria found especially in lungs and there is no evidence of C. burnetii
in heart and liver. This difference is closely related to the route of infection and the phase of infection in each
individual examined. These results indicate that it is possible that there is a difference in strain between C. burnetii
found in East Java and West Java, but still requires further examination.
