Exercise 2.3 (Replace with Extemporaneous Exercise)
Procyclidine hydrochloride IV 5 mg o.d. × 3/7
Morphine sulphate SC 1 mL stat.
Atropine sulphate SC 1 mL stat.
Procyclidine Injection B.P.C. 1973 MDV 40 mL
Procyclidine HCl 10 mg/2 mL 0.2 g
Chlorocresol 0.1 %w/v 20 mL (Stock 0.2 %w/v)
Water for injection qs ad 40 mL
Method
1. Weigh ingredients
2. Measure chlorocresol 20 mL (stock 0.02%w/v)
3. Dissolve active in water
4. Add chlorocresol 20 mL and mix the solution
5. Add water to 40 mL
6. Filter the solution using sintered glass filter grade 3
7. Check the solution using Allen viewer
8. Sterilise the solution by autoclave or filtration and transfer by aseptic technique to a
sterile container :
o Clean hands with soap and alcohol
o Clean laminar flow with alcohol
o Clean all the items with alcohol and arrange them in laminar flow
o Assemble the filtration unit:
▪ Cut wrapper of the syringe
▪ Take out plunger from the syringe
▪ Cut wrapper of the filter
▪ Join the filter with a syringe
▪ Cut wrapper of the bottle
▪ Place filter unit on the bottle
o Transfer 20 mL solution into syringe
o Push solution with plunger until all solution is transferred into the bottle
o Use forceps to take stopper
o Place rubber stopper on the bottle – close the bottle
o Place aluminum cap on the rubber stopper – use a crimper to tighten the aluminum cap
o Label preparation

Morphine sulphate 1 g 0.4 g
Atropine sulphate 0.06 g 0.024 g
Na metabisulphite 0.1 g 0.04 g
NaCl 0.69 g 0.276 g
*Chlorocresol 0.1 %w/v 20 mL (Stock 0.2 %w/v)
Water for injection qs ad 100 mL 40 mL
*Multidose vials: bactericide is necessary BPC pg 705-706
Method
1. Weigh all solids and dissolve accordingly. Measure chlorocresol.
2. Add WFI to 40 mL
3. Filter the solution using sintered glass filter grade 3
4. Check the solution using Allen viewer
5. Sterilise the solution heating with a bactericide or aseptic filtration.
