5-Partition chromatography (LLC)

partitioning of sample components between two immiscible
(partially miscible) phases.
Partition chromatography can be applied in TLC, paper & column
techniques

o The stationary phase is a liquid held on a suitable inert
       support

o The mobile phase is liquid (single solvent or mixture of
       solvents)
       the elution is isocratic

o The separation occurs as a result of differences in partition
       coefficient

o The Temperature must be kept constant during the process
       (it affects
       equilibrium)

Classification
According to the polarity of st.ph. and m. ph. It is classified to

1-Normal phase
Polar st.ph. & non polar m.ph.
the compounds are eluted in order of increasing polarity
(least polar solute eluted first).
2-Reversed phase
Non polar st.ph. & polar m.ph.
the compounds are eluted in order of decreasing polarity
(most polar solute eluted first).
