                  Aminoglycosides                                                                        30s

                   Streptomycin, Gentamicin, Tobramycin, Neomycin, Amikacin

                       •  Drugs derived from Streptomyces genus ……. suffix –mycin
                       •  Drugs derived from Micromonospora ………suffix -micin


                   Mechanism: irreversible binding to the 30S subunit causes
                       •  Inhibition of initiation
                       •   misreading of mRNA by ribosomes                               Bactericidal














                     • Poor oral absorption (very polar) => parenteral administration (IV, IM) and

                        topical application

                      • Excellent activity against aerobic Gram-negative bacteria
                         (Enterobacteriaceae and Pseudomonas aeruginosa)

                      •  Gentamicin has some activity against aerobic Gram-positive bacteria
                      •  Penetration into cell requires an oxygen-dependent transport => anaerobes

                         are resistant
                      •  No anaerobic or atypical coverage.

                      Toxicity

                         -Nephrotoxicity           -Ototoxicity

                    Dosage: one daily dose

                        → To decrease side effects
                        → Have concentration dependent killing
                        → Have post-antibiotic effect
