Isolation and purification of bioactive compound from the methanolic flower extracts
Leucas indica and evaluation of its anti-cancer activity using MTT assay

Abstract:
The present study was aimed to investigate the anti-cancerous properties of methanolic flower extracts
of Leucas indica was tested against the selected cancerous cell lines, HeLa, HCT116, HL-60 and MCF-
7 using MTT [(3-(4,5-dimethylthiazol-2-yl)-2,5- dipenyltetrazolium bromide)] assay. After isolation
and purification of compound from the methanolic flower extracts of L. indica using silica gel column
chromatography and TLC, physicochemical and spectroscopic analyses, e.g. UV, IR, 1 H NMR, 13 C
NMR, COSY, HSQC and mass spectroscopy were determined. The bioactive compound was identified
as a member that belongs to 4H-chromen-4-one. The findings suggested that the purified compound
showed cytotoxic effects against the selected cell lines using MTT assay. In addition, apoptotic cell
death was noticed in the cancerous cell lines treated with the bioactive compound of L. indica. It can be
concluded that the isolation and purification of L. indica flowers showed that the bioactive compound
belongs to 4H-chromen-one and the observation that the cytotoxic effects of the flowering parts of L.
indica against the selected cell lines may be suggestive of potential source in the development of natural
drugs for the treatment of cancer.
