Clotting Factor VIII is absent or deficient in the blood
of persons suffering from haemophilia A, i.e.
haemophilacs or “Bleeders “ .Concentrates of missing
protein are prepared from pooled normal plasma.
Fibrinogen being the principal constituent,and Factor
VIII
a trace component. For prolonged storage of the dried
product a temperature below -20 is desirable.
However for some weeks a temperature of 4-6⁰ is safe
and the product can be transported unrefrigerated
.Reconstitution is in water for injections at 20-30 ⁰ and
may be slow.The solution should be transfused
without delay as activity may be lost on standing.
2-Factor VIII concentrates may transmit Hepatitis B
3-Human Plasma Cryoprecipitate
A protein fraction containing Factor VIII is obtained by
freezing plasma & then thawing it under controlled
conditions .It can be made in transfusion centres &
blood
Banks without highly specialized facilities.
4-Factor VIII Concentrate Derived from Animal
Plasma
Made from bovine or porcine plasma .They have high
content of Factor VIII with a very high specific activity
After a limited period of treatment, the recipient
becomes sensitized to the foreign protein .Their use is
limited to life saving .They are labile &expensive &
their use should be controlled by assays carried out
