                                                     ABSTRACT

The 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) is lipophilic, has a low metabolism and excretion
rate, so it tends to accumulate in the body. The toxic effect of TCDD on the reproductive system is
suspected to reduce testosterone levels and suppress the growth and maturation of spermatozoa. In
addition, TCDD can destroy the antioxidant defences of the testicular chondrosomes and cytomicrosomes
systems, causing oxidative stress and affecting the reproductive ability of male animals α-tocopherol
known as vitamin E, is fat-soluble, effective as an antioxidant and immunomodulator and has an
antagonistic effect on endocrine toxicity and minimizes changes in testicular structure due to TCDD. This
study was conducted to determine the effect of -tocopherol on Leydig cell count and testosterone levels
in rats exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). A total of 50 rats were divided into five
groups, so that each group consisted of 10 rats that received treatment with TCDD and tocopherol. The
control group (C) was given TCDD of 0 ng/kg BW/day, and tocopherol was 0 mg/kg/day, group T0 was
given TCDD of 700 ng/kg BW/day, and tocopherol was 0 mg/kg BW/day, the T1 group was given TCDD
          of 700 ng/kg BW/day, and tocopherol was 77 mg/kg BW/day, group T2 was given TCDD of 700 ng/kg
BW/day, and tocopherol was 140 mg/kg BW/day, and group T3 was given TCDD as much as 700 ng/kg
BW/day and tocopherol as much as 259 mg/kg BW/day. Administration of TCDD and vitamin E for 45
days. The result indicated that the administration of vitamin E can maintain the number of Leydig cells
and testosterone levels in rats exposed to TCDD.
