Number Of Testic Sertoli Cells In Balb/C Male Mouses Exposed To Diazinone

ABSTRACT
Diazinon is an organophosphate class pesticide which is widely used in agricultural business. Diazinone
has cytotoxic and pathological effects that work in the body with the effect of inhibiting the acetylcholinesterase
enzyme, causing acetylcholine buildup in the synapse gap which can lead to cell apoptosis and hormonal
disturbances in the reproductive system. The purpose of this study was to determine the effectiveness of black
cumin seeds as an antioxidant to reduce the negative impact of free radicals in the body and to suppress caspase-
3 expression as a caspase mediator for apoptosis in the sertoli cells as a producer of substances needed in the
spermatogenesis process in the histopathological picture of the testes. This study was using male Balb / c mice
which were divided into 5 treatment groups, namely negatif control (K-) for untreated male mice; positive control
(K+) for male mice given diazinone 40 mg / kg BW for 8 weeks; whereas treatments 1, 2, and 3 (P1, P2, and P3)
were given exposure to diazinon 40 mg / kg BW for 8 weeks and black cumin extract 200, 400, and 600 mg / kg
BW at week 5-8, respectively. The results showed that giving black cumin seed extract for 3 weeks had an effect
on decreasing caspase-3 expression and increasing the number of sertoli cells.
